A Micromethod for the Determination of Uronic Acids*

چکیده
The original macromethod of Tollens and Lefevre (1) for the determination of uranic acids has undergone various modifications in the hands of different investigators (2--g). All of t,he modifications are based on the principle that the simple uranic acids and the complex polyuronide substances yield carbon dioxide quant.itatively when distilled with 12 per cent hydrochloric acid. In some the carbon dioxide is determined gravimetrically, in others by tit, rimetric methods. The size of the sample used varies between the limits of 0.2 to 1.0 gm. and the period of heating from 4 to 8 hours. Recently Buston (9) described a micromet.hod embodying titrimetric technique for the determination of uranic acid anhydride groups in pectic subst, ances. In conjunction with t,he studies on the carbonyl sugar acids in progress in this laboratory, one of us (K. P. L.) and a collaborator1 were engaged with the development of a satisfactory micromethod for their determination prior t,o the time of the appearance of Buston’s method. The details had not been completely worked out, consequently it appeared advisable to evaluate the merits a.nd defects of Buston’s method in conjunction with our work. Buston’s method is open to criticism. The apparatus is very

A new enzymatic method for the determination of free and conjugated glucuronic acid.
A new method is reported for the quantitative determination of glucuronic and galacturonic acid, which is based on spectrophotometric measurement of NADH. The NAD-linked oxidation of the uronic acids to the corresponding dicarboxylic acids is measured in the presence of uronic acid dehydrogenase. This enzyme was isolated from Pseudomonas syringae. The test is highly specific for glucuronic and ...
متن کاملDetermination of Uronic Acids in Cellulosic Materials
Methods for the determination of uronic acids, based on the estimation of the amount of carbon dioxide evolved by these acids in a given length of time during treatment with a boiling solution of hydrochloric acid, were found to fail when cellulose or other carbohydrate material was present, since these substances also decomposed with the evolution of some carbon dioxide. An investigation of th...

A method for simultaneous separation and quantitative determination of arabinose, galactose, glucose, xylose, xylonic acid, gluconic acid, galacturonic acid, and glucuronic acid was developed by using high performance anion-exchange chromatography coupled with pulsed amperometric detection (HPAEC-PAD). The separation was performed on a CarboPac TM PA-10 column (250 mm × 2 mm) with a various gra...
متن کاملPaper-chromatographic Separation of [3h]borohydride- Reduced Hexoses for Quantitative Determination of Polysaccharide Composition
An improved procedure for determination of composition of heteropolysaccharides by radiochromatography of sodium [3H]borohydride-reduced hydrolyzates has been developed. The method is accurate and sensitive, is less cumbersome and more reliable than similar procedures currently in use, and permits, for the first time, compositional analysis of hydrolyzates containing both hexoses and uronic aci...
متن کاملDifferential gas-liquid chromatography method for determination of uronic acids in carbohydrate mixtures.
An integrated gas-liquid chromatography method is described for the quantitation of mixtures containing simple monosaccharides in addition to mannuronic, glucuronic, and/or galacturonic acids. A hydrolyzed sample is divided into two portions. One portion is analyzed by the standard aldononitrile method. Glucuronic, galacturonic, and mannuronic acids are converted into compounds that do not chro...
